In the seventh round of the 2013 NFL Draft (218th overall), Poyer was chosen by the Philadelphia Eagles. Poyer and the Eagles agreed to a four-year, $2.22 million contract on May 9, 2013, which included a $60,256 signing bonus.

He went up against Brandon Boykin, Trevard Lindley, Curtis Marsh Jr., and Brandon Hughes for a reserve cornerback place during training camp.

After Cary Williams, Bradley Fletcher, and Brandon Boykin in the depth chart, Poyer was selected by head coach Chip Kelly as the fourth cornerback.

In the Philadelphia Eagles’ season-opening game against the Washington Redskins, he made his professional regular season debut. In the team’s 33–27 victory, he collected three total tackles.

After tight end Fred Davis received a ten-yard pass in the third quarter, he and teammate Nate Allen made his first career tackle. Shaun Prater and Roc Carmichael passed Poyer on the depth chart, and he was demoted to special teams.

After Poyer was a healthy scratch in the Philadelphia Eagles’ last two games (Weeks 5–6), the team dismissed him on October 19, 2013.

The Cleveland Browns claimed Poyer off waivers on October 21, 2013. Head coach Rob Chudzinski placed Josh Aubrey on injured reserve with knee and ankle problems, and Poyer was named the backup strong safety behind T.J. Ward upon Aubrey’s arrival.

On December 8, 2013, in a Week 14 loss to the New England Patriots, Poyer recorded a season-high six solo tackles. Following their 4–12 season, Cleveland Browns head coach Rob Chudzinski was sacked on December 30, 2013.

In 12 games and zero starts, he totaled 21 tackles (20 solo) by the end of his rookie campaign. In addition, he returned punts for 114 yards on eight occasions as a backup.

He faced competition from Darwin Cook, Josh Aubrey, Jim Leonhard, Johnson Bademosi, Robert Nelson, and others during training camp for a backup safety slot. To begin the regular season, head coach Mike Pettine selected Poyer as the backup free safety behind Tashaun Gipson.

In 2015, Poyer made a comeback to start the regular season as Tashaun Gipson’s backup free safety. Poyer made his first professional start on October 18, 2015, taking Gipson’s place after the latter had an ankle injury the week before. The Browns fell short of the Denver Broncos 26–23, despite his three solo tackles.

Poyer faced up against Rahim Moore throughout training camp to be the starting free safety following Tashaun Gipson’s departure via free agency. Poyer and Ibraheim Campbell were selected by head coach Hue Jackson to start as the starting free safety duo for the regular season.

Poyer signed a four-year, $13 million contract with the Buffalo Bills on March 9, 2017, which included a $3.50 million signing bonus and a $7.40 million guaranteed.

On the first day of free agency, he signed with the Bills right away, rejoining Bobby Babich, the former assistant defensive backs coach for the Cleveland Browns. During defensive backfield workouts at the 2013 NFL Scouting Combine, Babich first got to know Poyer.

Along with free safety Micah Hyde, head coach Sean McDermott selected Poyer as the starting strong safety to begin the regular season. With a 9–7 record, the Buffalo Bills placed second in the AFC East.

In his first-ever postseason game on January 7, 2018, Poyer recorded five total tackles in a 10-3 loss to the Jacksonville Jaguars in the Wild Card Round.

Poyer totaled 100 tackles, 4 interceptions, 2 sacks, and 1 forced fumble by the end of the 2018 campaign. Even though the Bills were eliminated from the playoffs in 2018, Poyer and Hyde’s defense held strong, concluding the season with the second-fewest passing yards allowed in the league.

Poyer and the Bills agreed to a two-year contract extension on March 19, 2020. Poyer received his first career selection to the 2021 All-Pro Team on January 14, 2022.

With the Bills finishing with the best defense in the league, he became the only NFL player to record at least five interceptions and three sacks in a single season by finishing the 2021 campaign with a career-high three sacks and tying the previous mark of five interceptions.

By his teammates, he came in at number 45 on the NFL Top 100 Players of 2022 list. In addition, he was placed 57th on the NFL Top 100 Players of 2023 list by his fellow players.

Poyer and the Bills agreed to a two-year contract extension on March 15, 2023. Poyer was released by the Bills on March 6, 2024.
