DeSantis, the son of Karen and Ronald Daniel DeSantis, was born in Jacksonville. He is of Italian ancestry: all of his eight great-grandparents were born in Italy and came from communes in the provinces of L’Aquila, Benevento, Avellino, and Campobasso.

His maternal great-great-grandfather Salvatore Storti arrived in the United States of America from Italy in 1904 and settled in Pennsylvania.

His great-great-grandmother Luigia Colucci traveled to America in 1917 along with her husband. His father set up Nielsen television rating boxes, and DeSantis’ mother was a nurse.
